Physical Map

A Chinese Chestnut (Castanea mollissima) physical map was developed from two BAC libraries. These libraries were constructed using partial digestions with EcoRI (10X coverage) and HindIII (11X coverage). For more information on the libraries, please click here.

BAC fingerprinting is based on high information content fingerprinting protocols (HICF) and standard capillary-based automated electrophoresis platforms/software. Contig analysis is being completed using FPC software.

Hybridizations are underway and, as well as anchoring the physical map to the genetic map, should help to merge contigs and provide more confidence in the assembly.

With an estimated average band size of 1150 bp, then total coverage of contigs is 1,261 Mb with the longest contig (#2560) covering 2944 kb. Assuming a genome of 794 kb and an average insert size of 119 kb, the physical map coverage is expected to be 18X.
